The authors reported two cases with severe COVID-19 and the IFIH1 putative loss-of-function variants c.1641 + 1G > C and c.2730C > A, which are likely to impair MDA5 activation, hence disrupting type I interferon activity and reducing viral clearance leading to severe COVID-19 outcomes.
