ifih1 loss of function predisposes to inflammatory and sars‐cov‐2‐related infectious diseases

  • What: The authors showed that six different IFIH1 variants were detected in nine patients suffering from severe COVID-19, MIS-C, periodic fever, inflammatory bowel disease, or immunodeficiency with glomerulonephritis. The authors reported two cases with severe COVID-19 and the IFIH1 putative loss-of-function variants c.1641 + 1G > C and c.2730C > A, which are likely to impair MDA5 activation, hence disrupting type I interferon activity and reducing viral clearance leading to severe COVID-19 outcomes.
