The authors used rat embryonic stem_cells (rESC) developed at Regeneron as the starting point for producing FRG rats (manuscript submitted). A plasmid-based DNA targeting vector was designed to delete the rat Il2rg gene by homologous recombination in rESC (Fig 1a-G). In separate experiments, Il2rg or Rag1/Rag2 mutant rESC clones were microinjected into host blastocyst-stage rat embryos . . .
