A high throughput screening assay to identify inhibitory antibodies targeting alphavirus release

HIGHLIGHTS

SUMMARY

Antiviral antibodies are most often screened for their ability to block viral entry (whether via attachment and internalization, or later fusion within acidified endosomes) but there is often little correlation between the potency of in_vitro entry neutralization and the ability to prophylactically and/or therapeutically protect in_vivo. Identification of NAbs that strongly inhibit budding by creating antibody/antigen complexes on the cell surface will also identify mAbs that will act potently through ADCC in_vivo. Lead candidate mAbs from these in_vitro screens can then be advanced to small animal models of CHIKV for more realistic . . .
