A validated hplc-diode array detection method for therapeutic drug monitoring of thiopurines in pediatric patients: from bench to bedside

HIGHLIGHTS

SUMMARY

Thiopurines, such as mercaptopurine (MP), its prodrug azathioprine (AZA) and thioguanine (TG), are part of the therapeutic armamentarium for the treatment of several pediatric diseases including inflammatory bowel disease (IBD) and acute lymphoblastic leukemia (ALL). The stock solutions of 100 mM TG and 100 mM MMP were prepared in DMSO and stored at -20 ◦ C. On the day of the analysis, working solutions of 20 µM TG and 100 µM MMP were prepared fresh from stock solutions by diluting, respectively, the analytes at 1:5000 and 1:1000 in HPLC grade water.
