The authors aimed to analyze published datasets of single-cell and bulk gene_expression in AD to learn more about two key basic questions regarding its pathogenesis: what are the cell type specific transcriptional alterations that are associated with it pathogenesis and progression and which cell-type is most transcriptionally altered? Interestingly, they found neurons to have more differentially expressed genes (DEGs) downregulated while other cell types such as astrocytes and microglia, had more genes upregulated, in accordance with the central role of neurons proposed by Frere and Slutstky.
