The cDNA of FASN truncation mutants was amplified from Flag-FASN by PCR and cloned into pcDNA3.1-Flag vector. HA-, and other DUBs were kept in the lab. Full length cDNAs of human FASN and , were also subcloned into the pLVX-neo vector. Plasmids expressing , shRNAs were constructed by annealing the sense and antisense oligonucleotides of the target sequence and cloned into the pLKO.1-puro vector.
