Episomes and transposases—utilities to maintain transgene expression from nonviral vectors

SUMMARY

    As outlined above, long-term nuclear maintenance and stable transgene_expression in therapeutically relevant cell types and tissues is a major challenge in nonviral_vector development. In this review, the authors will discuss recent progress in nonviral_vector development regarding (I) transgene_expression, (II) mitotic stability, and_(III) nuclear localization. The choice of the transgene promoter type has the most fundamental impact on transgene_expression levels. The cytomegalovirus (CMV) early enhancer/promoter facilitates a high level of transgene_expression in various cell types.
