SUMMARY
The pCMV6 plasmid with FLAG- and Myc-tagged human dopamine receptor D1 (DRD1) (Origene, cat# RC210389) was utilised for site-directed_mutagenesis using the QuikChange Lightning site-directed_mutagenesis kit (Stratagene, Santa Clara, CA, USA) with primers DRD1_T37K_F 50 -gttccccaggagcttggacaggatgagca-30 and DRD1_T37K_R 50 tgctcatcctgtccaagctcctggggaac-30. Untransfected HEK-293T_cells, cells transiently expressing either pCMV6-DRD1-WT or pCMV6-DRD1-T37K, and cells transfected with the pCMV6 empty vector, were washed with cold PBS and lysed in RIPA buffer (ThemoFisher, Massachusetts) plus protease inhibitor cocktail for 30 min at 4 ◦ C with inversion.
