Inhibition of insulin-like growth factors increases production of cxcl9/10 by macrophages and fibroblasts and facilitates cd8+ cytotoxic t cell recruitment to pancreatic tumours

  • What: The authors demonstrate an increase in aSMA+ CAFs upon IGF blockade . With regard to TAMs, no changes in total F4/80+ TAM numbers are observed . As a positive control the authors demonstrate a significant increase in CD8+ T_cell migration towards RPMI upon the addition of recombinant CXCL9 (1000 ng/ml) (Supplementary Figure S2J). 3.5 IGF blockade leads to increased CD8+ T_cell recruitment towards human PDAC tumours Having demonstrated that blocking IGF signalling increases the recruitment/trafficking of CD8+ T_cells towards the PDAC TME via CXCL9/10 in both in_vitro assays and in a mouse PDAC . . .
