Labeling of methyl groups: a streamlined protocol and guidance for the selection of 2h precursors based on molecular weight

  • What: The authors show that it is possible to obtain 2D 1H-13C spectra of a 50 kDa protein that are suitable for resonance assignment purposes by growing the cells in the presence of 100% ­H2O and protonated glucose, with the only source of deuterium from highly deuterated 13C precursors. The work indicates that the synthesis of similarly labeled Val and Ile would also be advantageous in those systems.
